Von 1968 bis 1972 besuchte er die Architectural Association School London. Koolhaas war Gaststipendiat am Institut für Architektur und Stadtbau in New York. Er gründete 1975 das Office for Metropolitan Architecture [OMA] mit Elia und Zoe Zenghelis und Madelon Vriesendorp in London. Seither ist er in der Gebäude- und Städteplanung tätig. Seine Interessensgebiete erstrecken sich über zeitgenössische Gesellschaft, Gebäude und städtebauliche Entwicklung. Seit 1995 ist er Professor für Architektur und Stadtdesign an der Harvard Universität. Er betreibt Studien zu Design als eine gezielte Untersuchung über derzeitige urbane und architektonische Bedingungen in verschiedenen Erdteilen. Zur Zeit ist Koolhaas mit dem »Project on the City« beschäftigt.
